Common Applications
- Ultrastructural and morphological studies of biological samples
- Negative staining of virus particles, bacteria, and protein complexes
- Immunogold labeling for macromolecule localization
- High-resolution imaging of thin-sectioned specimens
Services
Ultra-Thin Sectioning for Morphology
Biological specimens are fixed, dehydrated, and embedded in plastic blocks, then sectioned into approximately 80 nm sections. After staining with electron-dense stains, sections are observed under the electron microscope. A standard fixation and embedding protocol is used for optimal ultrastructural preservation. Custom protocols are available — contact us to discuss.
Negative Staining
A technique for visualizing particulate biological samples in suspension — including virus particles, bacteria, bacteriophages, and protein or lipoprotein complexes. An electron-dense stain such as uranyl acetate or lead citrate is applied around the specimen, which is then placed onto formvar carbon-coated grids for observation.
Immunogold Labeling
A powerful technique providing information on the identity, localization, and distribution of macromolecules at the ultrastructural level. The best preparative method is worked out individually for each experimental system — contact us to discuss your project before starting.
